Absolutely.  Some of the coolest bands out there are the ones who "break the rules."

But you combined your term "preference" with the statement "rock bands must have a bassist."  That sounds more like a rule to me.  A rule that was successfully broken by (hereigoagain) the Doors (and plenty others I'm sure).  :P
